Abstract
Staphylococcal nuclease has been modified by reaction with an excess of diketane. Spectroscopic and immunological features of the native protein and of the derivative have been compared. Unlike acetylation or trifluoroacetylation, acetoacetylation of the lysine residues, has been found to produce a marked disorganization in the conformation of the enzyme. The modified enzyme shows a new absorption band, not present in simple model compounds. Removal of blocking groups from the modified enzyme by hydroxylamine is not complete.
