Abstract
Optical absorption spectroscopy in the temperature range 300 10 K is shown to be an useful experimental tool for studies of hemeprotein dynamics; in particular, the temperature dependence of the gaussian broadening of an absorption band gives information on the “bath” of protein soft modes () coupled to the electronic transition involved. Two different dynamic regimes are evidenced with this technique: at low temperatures each protein is frozen in a given conformational substate and only harmonic nuclear vibrations are detected; at higher temperatures anhaimonic contributions to soft modes are evident: these are attributed to the onset of fluctuations among different conformational substates, The switch between these two dynamic regimes is shown to depend upon protein tertiary structure and state of ligation, as well as on the composition of the solvent matrix. The relevance of dynamic properties to hemeproteins functionality is also discussed.
