Abstract
Curcumin and genistein are two natural products obtained from Curcuma longa L. and soybeans, endowed with many biological properties. Recently, both curcumin and genistein were proven to possess an antitrypanosomal activity: curcumin was active against Trypanosoma brucei strain 427 with an EC50 values of 2.5 μM, while genistein was effective against Trypanosoma brucei rhodesiense, with an EC50 value of 4.81 μM. Thus, with the aim to identify the target protease of both natural products, in the present study we investigated the in vitro effects of curcumin and genistein, alone and in combination, against rhodesain, the main cysteine protease of Trypanosoma brucei rhodesiense, which is responsible for the acute form of Human African Trypanosomiasis (HAT). Rhodesain is required for the parasite to cross the blood brain barrier of the human host, thus causing the late (neurological) stage of the disease; it is also involved in the turnover of variant surface glycoproteins (VSGs) of trypanosomes and in the degradation of host immunoglobulins, thus evading the host immune responses. In addition to these functions, rhodesain is a lysosomal protease, essential for parasite survival, thus being considered the primary target for HAT treatment.
